چکیده

Hyperoxia is known to injure tissues and cells (1) . In a series of papers the quantitative influence of hyperoxia on some aspects of lung ultrastructure was elucidated (2-4) . These studies revealed that hyperoxia resulted in rapid destruction of the pulmonary capillaries, a thickening of the air-blood barrier, and, eventually, a repopulation of the alveolar epithelium by granular pneumocytes at the expense of the type I alveolar cell . While studying the interrelation between protein synthesis by the lung and the ultrastructure of the pulmonary granular pneumocyte under hyperoxic conditions (5, 6), we noted that the mitochondria of granular pneumocytes from 02-exposed rats seemed to contain fewer granules than those from rats exposed to compressed air . This led to the present study in which we used stereological techniques to examine the influence of in vivo hyperoxia on the dense granule content of mitochondria of rat lung granular pneumocytes .

Hyperoxia: a stereologic ultrastructural examination of its influence on cytoplasmic components of the pulmonary granular pneumocyte.

We used the technique of lineal analysis to study the influence of 48 h of hyperoxia on cytoplasmic organelles of pulmonary granular pneumocytes with particular reference to their lamellar bodies. We undertook this study because lamellar bodies are considered to be storage granules for pulmonary surfactant and because we had found that hyperoxia decreased [(14)C]leucine incorporation into prote...

Adaption to hyperoxia. Influence on protein synthesis by lung and on granular pneumocyte ultrastructure.

We studied the influence of prolonged exposure to hyperoxia (O(2) > 98%) on protein synthesis and on the ultrastructure of the granular pneumocyte. To study protein synthesis, as indicated by l-[U-(14)C]-leucine incorporation into protein, lung slices were incubated with radioactive leucine and a surface-active fraction was obtained by ultracentrifugation of lung homogenates. We found that, fol...

Inhibition of TNFalpha in vivo prevents hyperoxia-mediated activation of caspase 3 in type II cells

BACKGROUND The mechanisms during the initial phase of oxygen toxicity leading to pulmonary tissue damage are incompletely known. Increase of tumour necrosis factor alpha (TNFalpha) represents one of the first pulmonary responses to hyperoxia. We hypothesised that, in the initial phase of hyperoxia, TNFalpha activates the caspase cascade in type II pneumocytes (TIIcells). METHODS Lung sections...

Ultrastructural study of naturally occuring ovine pulmonary adenocarcinoma in Fars province, Iran

Ovine pulmonary adenocarcinoma (OPA) was studied in the lungs of 15 naturally-affected sheep (9 with classical and 6 atypical lesions) by transmission electron microscopy. Two lung samples from normal sheep were used to develop the ultrastructural criteria. Pathologic lesions consisted of focal-multifocal to coalescent nodules or masses in the cranioventral or diaphragmatic lobes. Ultrastructur...
